Easiest way for me to get Tomahawk Mastery rank 2 in Red Dead Redemption was to head to Tesora Azul, ride up on your horse and throw a tomahawk at the first guy guarding the gate. As soon as you kill him hop off your horse and throw a tomahawk at the first guy that comes running out of the gate, I hid behind the wagon to recover before I attacked the third guy that comes running out. Just run up to him and melee kill him with the tomahawk.
